Activated Protein C Resistance

Overview

  • EPIC Code:
  • LAB2559
  • Soft Test Code:
  • APCR
  • Send Out Test Code:
  • 22
Alternate Names
  • Act Protein C Resist
  • Activated Protein C
  • Activated Protein C Resistance
  • APC Resistance
  • APC Resistance, APCR
  • APC-R
Clinical Significance

Activated protein C (APC) resistance is the most frequent hereditary defect associated with deep vein thrombosis. Over 95% of the APC resistance phenotype is due to the Factor V Leiden mutation. The APC-Resistance assay is a plasma based functional test for the determination of APC resistance caused by the factor V Leiden mutatio

Specimen Collection & Preparation

Specimen Requirements:

-OR-

2.0 mL Plasma from a Blue Top Tube - 3.2% Buffered Sodium Citrate in a Plastic Vial
Minimum Volume:
1.0 mL Na Citrate Plasma*

* This volume does not allow for repeat testing

Processing Instructions:

Centrifuge light blue-top tube for 15 minutes at approximately 1500 x g within 60 minutes of collection.

Using a plastic pipette, remove plasma, taking care to avoid the WBC/platelet buffy layer and place plasma into a plastic vial.

Centrifuge a second time and transfer platelet-poor plasma into a new plastic vial.

Plasma must be free of platelets (< 10,000/mcL).

Rejection Criteria:

Ambienty Specimen

Refrigerated Specimen

Clotted specimen

Serum

Gross hemolysis

Transport and Storage:
  • Frozen (-20° C or colder): 60 Days

Freeze immediately and ship on dry ice.

Clinical Interpretation

Reference Range:

≥2.1 Ratio

Methodology:
  • RVVT Based Assay (Clot-based)
