Information regarding flow cytometry. The process involves a specimen-usually blood, by can be a piece of tissue like a lymph node, fluids, and bone marrow. The specimen is lysed to get rid of the red cells. Then it is added to specific antibodies that help the pathologist to determine if the patient has a lymphoma or leukemia. We also process peripheral blood to look for immunodeficiency, and HIV patients to check their T cell status.

 

The specifics:

 

CD4 (Tcell) and Lymph subset- are done on peripheral blood collected in a Sodium heparin (dark green with stripes on label) tube and kept at room temperature. These tests are batched and run on Monday, Wednesday, Friday, and Saturday

 

Acute Panels- are usually performed when blasts cells are present peripherally and/or a known leukemic patient for staging. This panel can be done on peripheral blood, but Bone Marrow is the preferred specimen.  Specimens should be collected into Sodium Heparin (Dark Green with stripes on label) tube and kept at room temperature. (Alternate specimen is Lavender tube at room temperature)

 

B-cell Clonal Panel - Peripheral blood or Bone Marrow can be used. Collect in Sodium Heparin (Dark Green with stripes) and kept at room temperature. (Alternate specimen is Lavender tube at room temperature)

 

Tissue- This is ordered by the pathologist for a tissue sample, like a lymph node or mass that has been collected in surgery. These specimens are put in RPMI, a culture media, to preserve it until testing is done. These samples are kept refrigerated until tested.

 

Fluids- No Additive Tube or EDTA (lavender) tube. Fluids should be refrigerated until tested. Complete panels can be done on fluids.

 

Bronchial lavage fluids- CD4/CD8 ratio: All BAL specimens will automatically reflex CD4/CD8 ratio when the lymphocyte count is > 15%.
