Clinical Significance

The T-SPOT®. TB test is an in vitro diagnostic test for the detection of effector T cells that respond to stimulation by Mycobacterium tuberculosis antigens ESAT-6 and CFP 10 by capturing interferon gamma (IFN-g) in the vicinity of T cells in human whole blood collected in lithium heparin. It is intended for use as an aid in the diagnosis of M.tuberculosis infection. The T-SPOT®. TB test is an indirect test for M.tuberculosis infection (including disease) and is intended for use in conjunction with risk assessment, radiography and other medical and diagnostic evaluations.

Client Notes

Specimen Requirements may be different based on patients WBC (White Blood Cell Count).

Contact Client Response at 266-1500 (Option 1) for assistance if the patients WBC count is below normal Range.

If Patients has a low WBC, please notify the laboratory with a comment on the order “low WBC, please collect 2 extra vials for Tspot testing”


Specimen Overview

Due to the short stability of the specimen, and the needed time to get to the reference laboratory.

Specimen should only be collected Monday thru Friday

Specimen should not be collected before 6 a.m.

Specimen Requirements:

Adults and children ≥10 years: 6.0 mL Whole Blood in a Green Top Li Hep (9.0 mL Greiner) Tube
Minimum Volume:
6.0 mL Whole Blood*

* This volume does not allow for repeat testing

Processing Instructions:

Do not Centrifuge 

A patient’s cells can be pooled, if necessary to obtain sufficient cells from multiple tubes of blood which were collected and processed concurrently


Rejection Criteria:

Hemolysis

Anticoagulants other than lithium or sodium heparin

Centrifuged


Transport and Storage:
  • Ambient (18-24°C): 54 hours

    Transport

  • Refrigerated (2-8°C): Unacceptable
  • Frozen (-20° C or colder): Unacceptable
